Repeated granulation of antazoline-HCl (I) [2508-72-7] with 5% and 10% Et cellulose [9004-57-3] and shellac solutions, followed by release-time study of the drug from its tablets, showed no increase in the release proportional to the concentration of the former binder. However, with shellac the increase in the release time was proportional to the concentration increase. In both cases, the release of I was relatively more rapid in gastric fluid than in that observed in intestinal fluid. The effects of number of granulations and surface area of the tablet on drug release were also studied. Shellac was more effective than Et cellulose in reducing the release of the drug.

Addition of Mg stearate [557-04-0] and hydrogenated castor oil to antazoline-HCl (I) [2508-72-7] tablets made with Et cellulose [9004-57-3], prolonged the drug release. The tablets containing 15% and 20% Mg stearate released I more slowly in the gastric fluid than in the intestinal fluid, the release being more pronounced with the latter. Hydrogenated castor oil (10-20%) was perferred since it caused a gradual and uniform drug release both in gastric and intestinal fluids. Of the various retarding materials studied, hydrogenated castor oil caused the least erosion of I tablets, followed by Mg stearate.

Diphenhydramine-HCl, antazoline-HCl and chlorpheniramine maleate did not affect the stability of the dye. On the other hand, ascorbic acid caused fading of the dye solution within few hours. Antazoline-HCl was incorporated into a solid agar medium in order to study the isolation and identification of certain gram-neg. bacteria from stool samples. The medium for isolation of gram-neg. bacteria from gram-pos. bacteria contained 0.625 mg/ml of antazoline-HCl (called ALA-medium A), while the medium for isolating non-lactose fermenting gram-neg. bacteria contained 0.7 mg/ml of antazoline-HCl (called ALA-medium B). A comparison was made of the inhibitory action on different microorganisms of various antazoline-HCl concentrations and MacConkey’s agar and SS agar. Non-lactose fermenters were successfully isolated from stool samples before and after enrichment in selenite broth.

Allergic rhinitis, acute nasal congestion and sinusitis are one of the most common health problems and have a major effect on the quality of life. Several medications are used to improve the symptoms of such diseases in humans. Pharmaceutical pomade form containing Ephedrine (EPD) HCl, Naphazoline (NPZ) HCl, Antazoline (ANT) HCl, and Chlorobutanol (CLO) is one of them. Objective: For these reasons, this study includes the development of spectrophotometric and chromatog. methods for the determination of EPD HCl, NPZ HCl, ANT HCl, and CLO active agents in the pharmaceutical pomade. Method: In the spectrophotometric method, third-order derivative of the amplitudes at 218 nm n=5 and the first-order derivative of the amplitudes 254 nm n=13 was selected for the determination of EPD, ANT, resp. while NPZ was determined by the second derivative at 234 nm and n=21. Colorimetric detection was applied for assay anal. of CLO at 540 nm. Furthermore, a reverse phase high performance liquid chromatog. (RP- HPLC) method has been developed and optimized by using Agilent Zorbax Eclipse XDB C18 (75 mm x 3.0 mm, 3.5μm) column. The column temperature was 40°C, binary gradient elution was used and the mobile phase consisted of 15 mM phosphate buffer in distilled water (pH 3.0) and methanol, and the flow rate was 0.6 mL min-1 and the UV detector was detected at 210 nm. The linear operating range was obtained as 11.97-70, 0.59-3, 2.79-30, and 2.92-200μg mL-1 for EPD HCl, NPZ HCl, ANT HCl, and CLO resp. The LOD values were found to be 3.95, 0.19, 0.92 and 0.96μg mL-1 for EPD HCl, NPZ HCl, ANT HCl, and CLO in the spectrophotometric method, resp. The linear ranges in the RP-HPLC method were 8.2-24.36μg mL-1, 0.083 – 0.75μg/mL, 2.01-7.5μg mL-1 and 2.89-24.4μg mL-1 for EPD HCl, NPZ HCl, ANT HCl, and CLO, resp. The LOD values in the validation studies were 2.7, 0.025, 0.66 and 0.86μg mL-1 for EPD HCl, NPZ HCl, ANT HCl, and CLO in RP-HPLC method resp. Conclusion: The results of the spectrophotometric and chromatog. methods were compared and no differences were found between the two methods.

In order to evaluate the pharmacokinetics characteristic of antazoline hydrochloride in Beagle dogs, a sensitive and specific HPLC method was developed and validated using phenacetin as the internal standard (IS). The analyte and the IS were extracted from dog plasma by Et acetate under the basic condition. The analyte was separated by a C18 column and detected with a variable wavelength UV-detector. The mobile phase consisted of methanol-5 mmol L-1 tetra-Bu ammonium bromide (45:55, volume/volume) containing 0.5% glacial acetic acid in a flow rate of 1.0 mL min-1. Standard calibration graph for antazoline was linear over a curve range of 20-1600 ng mL-1 (R > 0.99) and the lower limit of quantification was 20 ng mL-1 using a plasma sample of 500 μL. The intra- and inter-day precision values were less than 14.3% relative standard deviation (RSD). The intra-day assay accuracy was in the range of 98.1-100.6% and the inter-day assay accuracy in the range of 99.2-101.1%. The extraction recoveries were on the average of 88.4% for antazoline and 76.8% for IS. Plasma samples were stable at least for 1 mo at -20 °C. This method was successfully applied to pharmacokinetics study of antazoline after i.v. administration to Beagle dogs.

A reversed-phase ion pair chromatog. method with liquid-liquid extraction anal. method was developed and validated for the determination of antazoline hydrochloride in plasma and excreta of rat. The aim of our study was to characterize the preclin. pharmacokinetics and excretion profiles of antazoline hydrochloride in rats after i.v. injection at the dose of 10 mg/kg. Plasma and excreta samples were extracted with Et acetate, and phenacetin was used as the internal standard The result showed that the method is suitable for the quantification of antazoline hydrochloride in plasma and excreta samples. Anal. of accuracy (90.89-112.33%), imprecision (<7.1%) and recovery (>82.5%) showed adequate values. After a single i.v. administration at 10 mg/kg to rats, plasma concentration profile showed a relative fast elimination proceeding with a terminal elimination half-life of 3.53 h. Approx. 61.8 and 14.2% of the administered dose were recovered in urine and bile after 72 and 24 h post-dosing resp.; 5.9% of the administered dose was recovered in feces after 72 h post-dosing. NMR anal. showed that in the 1:1 complexes the total antazoline fraction was present as an inclusion complex, whereas tetracaine was only partly included in spite of a similar phase solubility diagram.
