Cozzolino, Alessia research published 《 [“Block and replace” of sodium levothyroxine therapy].》, the research content is summarized as follows. We present the case of a 65 years old patient with Graves disease associated with hyperthyroidism and a medical history of bipolar disorder treated with lithium. Hyperthyroidism was initially treated with high-dose methimazole monotherapy and later the patient developed overt hypothyroidism (clinical and biochemical), but without remission of the underlying autoimmune disease. A “block and replace” therapeutic regimen was then started with reduced methimazole doses in combination with levothyroxine, which resulted in a short time in normalization of the hormonal profile and significant improvement of the clinical picture. Therefore, the “block and replace” regimen represents a valid therapeutic alternative to anti-thyroid drugs monotherapy in the treatment of hyperthyroidism due to Graves disease in selected cases.

Urease plays a key role in urinary tract and gastrointestinal infections for which inhibition of it could prove to be substantial as a potential therapeutic option. Here, synthesized thirty novel compounds in four different series based on methimazole skeleton, also known as thiamazole, a drug that is used for the treatment of hyperthyroidism. Characterization of novel compounds consisting oxadiazoles, hydrazine-1-carbothioamides and N′-arylidenebenzohydrazides were performed by 1H-, 13C-NMR and high resolution mass spectrometry (HRMS). The synthesized mols. were examined for structure-activity relationship in urease inhibition and found to exhibit inhibitory potency in a range of IC50 =3.56-14.08 μM compared to urea and acetohydroxamic acid used as standard Moreover, mol. docking was performed to understand ligand binding interactions in the active site of the enzyme (Jack bean urease, Protein Data Bank ID 3LA4). The promise of addressing urinary tract and gastrointestinal disorders through urease inhibition is discussed.

K. research published 《 Chlamydia pneumoniae can infect the central nervous system via the olfactory and trigeminal nerves and contributes to Alzheimer’s disease risk》, the research content is summarized as follows. Abstract: Chlamydia pneumoniae is a respiratory tract pathogen but can also infect the central nervous system (CNS). Recently, the link between C. pneumoniae CNS infection and late-onset dementia has become increasingly evident. In mice, CNS infection has been shown to occur weeks to months after intranasal inoculation. By isolating live C. pneumoniae from tissues and using immunohistochem., we show that C. pneumoniae can infect the olfactory and trigeminal nerves, olfactory bulb and brain within 72 h in mice. C. pneumoniae infection also resulted in dysregulation of key pathways involved in Alzheimer’s disease pathogenesis at 7 and 28 days after inoculation. Interestingly, amyloid beta accumulations were also detected adjacent to the C. pneumoniae inclusions in the olfactory system. Furthermore, injury to the nasal epithelium resulted in increased peripheral nerve and olfactory bulb infection, but did not alter general CNS infection. In vitro, C. pneumoniae was able to infect peripheral nerve and CNS glia. In summary, the nerves extending between the nasal cavity and the brain constitute invasion paths by which C. pneumoniae can rapidly invade the CNS likely by surviving in glia and leading to Aβ deposition.

Methimazole is a commonly used drugs for treatment of hyperthyroidism. Researching the solubility property of methimazole in multifarious solvent systems can be used for optimizing the purification process of methimazole in industry. In this study, the solubility properties of methimazole in twelve pure solvents (methanol, ethanol, 1-propanol, isopropanol, 1-butanol, isobutanol, 1,4-dioxane, Me acetate, acetonitrile, Et acetate, THF, acetone) and four binary mixed solvents (methanol + Et acetate, methanol + acetonitrile, methanol + isopropanol, methanol + Me acetate) were tested within 278.15 K to 323.15 K under 101.325 kPa. The results show that methimazole has the highest solubility in methanol and the smallest solubility in Et acetate. In addition, solubility is pos. correlated with temperature in pure solvents and mixed solvents. The XRD spectrum results show that the crystal form of methimazole before and after dissolving in twelve pure solvents do not change. The DSC test shows that the m.p. of methimazole is 417.27 K, and it is also proved that methimazole do not decompose before reaching the m.p. Herein, the Modified Apelblat model and Buchowski-Ksiazaczak λh model were used to fit the solubility data of methimazole in pure solvents. Jouyban-Acree model, CNIBS/R-K model and SUN model were used to fit the solubility data of methimazole in binary mixed solvents. The results show that Modified Apelblat model and CNIBS/R-K model have the best correlation with the solubility of methimazole. Anal. using the KAT-LSER model reveals that the influence of the interaction between part of the solvent and solute mol. on solubility The results show that the impact of dipolarity/polarizability is dominant.

Busby, Robert W.;Cai, Xiaokun;Yang, Sihyung;Ramos, Luis;Venkatarangan, Lata;Shen, Helen;Wax, Stephen;Sadeque, Abu J. M.;De Colle, Cyril research published 《 Metopimazine is primarily metabolized by a liver amidase in humans》, the research content is summarized as follows. Metopimazine (MPZ) is a peripherally restricted, dopamine D2 receptor antagonist used for four decades to treat acute nausea and vomiting. MPZ is currently under clin. investigation for the treatment of gastroparesis (GP). MPZ undergoes high first-pass metabolism that produces metopimazine acid (MPZA), the major circulating metabolite in humans. Despite a long history of use, the enzymes involved in the metabolism of MPZ have not been identified. Here we report a series of studies designed to identify potential MPZ metabolites in vitro, determine their clin. relevance in humans, and elucidate the enzymes responsible for their formation. The findings demonstrated that the formation of MPZA was primarily catalyzed by human liver microsomal amidase. Addnl., human liver cytosolic aldehyde oxidase (AO) catalyzes the formation of MPZA, in vitro, although to a much lesser extent. Neither cytochrome P 450 enzymes nor flavin-monooxygenases (FMO) were involved in the formation MPZA, although two minor oxidative pathways were catalyzed by CYP3A4 and CYP2D6 in vitro. Anal. of plasma samples from subjects dosed 60 mg of MPZ verified that these oxidative pathways are very minor and that CYP enzyme involvement was negligible compared to microsomal amidase/hydrolase in overall MPZ metabolism in humans. The metabolism by liver amidase, an enzyme family not well defined in small mol. drug metabolism, with minimal metabolism by CYPs, differentiates this drug from current D2 antagonists used or in development for the treatment of GP.

Cai, Yun;Wang, Zhixiao;Liu, Xiaoyun;Wei, Ling;Li, Shushu;Zheng, Xuqin;Yang, Tao;Xu, Xinyu research published 《 The frequency of intrathyroidal follicular helper T cells varies with the progression of Graves’ disease and Hashimoto’s thyroiditis》, the research content is summarized as follows. Autoimmune thyroid diseases (AITD), mainly Graves’ disease (GD) and Hashimoto’s thyroiditis (HT), are common organ-specific autoimmune diseases characterized by circulating antibodies and lymphocyte infiltration. Follicular helper T (Tfh) cell dysregulation is involved in the development of autoimmune pathologies. We aimed to explore the role of intrathyroidal and circulating Tfh cells in patients with GD and HT. Ultrasound-guided thyroid fine-needle aspiration (FNA) was conducted in 35 patients with GD, 40 patients with HT, and 22 patients with nonautoimmune thyroid disease (nAITD). Peripheral blood samples were also obtained from 40 patients with GD, 40 patients with HT, and 40 healthy controls. The frequencies of intrathyroidal and circulating Tfh cells from FNA and peripheral blood samples were assessed by flow cytometry. Addnl., the correlations between the frequencies of the Tfh cells and the levels of autoantibodies and hormones or disease duration were analyzed. The frequency of intrathyroidal CD4+CXCR5+ICOShigh Tfh cells was higher in HT patients than in GD patients. Significant correlations were identified between the percentages of circulating and intrathyroidal Tfh cells and the serum concentrations of thyroid autoantibodies, especially thyroglobulin antibodies (TgAb), in AITD. Intrathyroidal CD4+CXCR5+ICOShigh Tfh cells were pos. correlated with free triiodothyronine (FT3) in HT patients but neg. correlated with FT3 in GD patients. In addition, HT patients with a longer disease duration had an increased frequency of intrathyroidal CD4+CXCR5+ICOShigh and CD4+CXCR5+PD-1+ Tfh cells. In contrast, in the GD patients, a longer disease duration did not affect the frequency of intrathyroidal CD4+CXCR5+ICOShigh but was associated with a lower frequency of CD4+CXCR5+PD-1+ Tfh cells. Our data suggest that intrathyroidal Tfh cells might play a role in the pathogenesis of AITD and they are potential immunobiomarkers for AITD.

Cantiello, Michela;Carletti, Monica;Giantin, Mery;Gardini, Giulia;Capolongo, Francesca;Cascio, Paolo;Pauletto, Marianna;Girolami, Flavia;Dacasto, Mauro;Nebbia, Carlo research published 《 Induction by Phenobarbital of Phase I and II Xenobiotic-Metabolizing Enzymes in Bovine Liver: An Overall Catalytic and Immunochemical Characterization》, the research content is summarized as follows. In cattle, phenobarbital (PB) upregulates target drug-metabolizing enzyme (DME) mRNA levels. However, few data about PB’s post-transcriptional effects are actually available. This work provides the first, and an almost complete, characterization of PB-dependent changes in DME catalytic activities in bovine liver using common probe substrates and confirmatory immunoblotting investigations. As expected, PB increased the total cytochrome P 450 (CYP) content and the extent of metyrapone binding; moreover, an augmentation of protein amounts and related enzyme activities was observed for known PB targets such as CYP2B, 2C, and 3A, but also CYP2E1. However, contradictory results were obtained for CYP1A, while a decreased catalytic activity was observed for flavin-containing monooxygenases 1 and 3. The barbiturate had no effect on the chosen hydrolytic and conjugative DMEs. For the first time, author also measured the 26S proteasome activity, and the increase observed in PB-treated cattle would suggest this post-translational event might contribute to cattle DME regulation. Overall, this study increased the knowledge of cattle hepatic drug metabolism, and further confirmed the presence of species differences in DME expression and activity between cattle, humans, and rodents. This reinforced the need for an extensive characterization and understanding of comparative mol. mechanisms involved in expression, regulation, and function of DMEs.

Bagur, Sophie;Lefort, Julie M.;Lacroix, Marie M.;de Lavilleon, Gaetan;Herry, Cyril;Chouvaeff, Mathilde;Billand, Clara;Geoffroy, Helene;Benchenane, Karim research published 《 Breathing-driven prefrontal oscillations regulate maintenance of conditioned-fear evoked freezing independently of initiation》, the research content is summarized as follows. Abstract: Brain-body interactions are thought to be essential in emotions but their physiol. basis remains poorly understood. In mice, regular 4 Hz breathing appears during freezing after cue-fear conditioning. Here we show that the olfactory bulb (OB) transmits this rhythm to the dorsomedial prefrontal cortex (dmPFC) where it organizes neural activity. Reduction of the respiratory-related 4 Hz oscillation, via bulbectomy or optogenetic perturbation of the OB, reduces freezing. Behavioral modeling shows that this is due to a specific reduction in freezing maintenance without impacting its initiation, thus dissociating these two phenomena. dmPFC LFP and firing patterns support the region′s specific function in freezing maintenance. In particular, population anal. reveals that network activity tracks 4 Hz power dynamics during freezing and reaches a stable state at 4 Hz peak that lasts until freezing termination. These results provide a potential mechanism and a functional role for bodily feedback in emotions and therefore shed light on the historical James-Cannon debate.

An efficient and practical method for the C-S cross coupling of thiophenols with aryl iodides using a Cu(I) catalyst was reported. A diverse set of thiophenols was coupled with electron rich and poor aryl iodides to obtain diaryl sulfides in good to excellent yields. Noteworthily, these reactions proceeded smoothly in polar protic solvents and under ligand-free environments. This procedure also finds application in the synthesis of 2-aminophenyl sulfide derivatives via ring opening of readily available benzothiazole. Furthermore, performed kinetics and computational studies to understand the mechanism of the Cu(I) catalyzed reaction in the absence of a ligand.

Sudheer research published 《 Photodegradation of 5-flurouracil, carvedilol, para-chlorophenol and methimazole with 3D MnWO4 nanoflower modified Ag2WO4 nanorods: A non-genotoxic nanomaterial for water treatment》, the research content is summarized as follows. The present study focused on the photocatalytic degradation of 5-Flurouracil (FU), carvedilol (Car), para-chlorophenol (PCP) and methimazole (Met) under visible light irradiation by MnWO4/Ag2WO4 (MWO/AWO) nanohybrid. Here, MWO/AWO nanohybrid was characterized by XRD, TEM, EDS, XPS, ESR, EIS, BET and DRS. The band gap energy of the MWO/AWO nanohybrid was found to be 2.75 eV, which enables effective photocatalytic activity of nanohybrid under visible light. The photocatalytic degradation of various PhACs such as Fu, Car, PCP and Met was found to be 98.8, 100, 98 and 98.1% resp. The degradation efficiency of the MWO/AWO nanohybrid on various PhACs was higher than the pure MWO and AWO nanoparticle. The effective formation of OH• and •O2 by MWO/AWO nanohybrid played an important role in degradation of PhACs and it was determined by radical scavenging experiment Further, the intermediates formed during the photocatalytic process were analyzed by GC-MS/MS to elucidate the photodegradation pathway and the results reveal the complete mineralization of the PhACs. The toxicity of the degraded product was performed against on Bacillus subtilis and Escherichia coli where it shows that the nanohybrid possesses high relative growth inhibition than AWO and MWO nanoparticles. In addition, the genotoxicity of the nanohybrid against Allium cepa was performed and it exhibited lower toxicity. The synthesized nanohybrid proves to be an excellent photocatalyst with good stability, reusability, eco-friendly, and cost-effective material for implementation in practical applications.

Application of C4H6N2S, Methimazole is an antithyroid compound found to have antioxidant properties. Methimazole inhibits activation of the IFN-g-induced Janus kinase (JAK)/STAT signaling pathway in FRTL-5 thyroid cells, which may account for its immunodolulatory effects. Additionally, methimazole is an inhibitor of thyroperoxidase.

Methimazole is a thiourea antithyroid agent that prevents iodine organification, thus inhibiting the synthesis of thyroxine. Antihyperthyroid.

Methimazole is an inhibitor of thyroid hormone synthesis. It is a substrate for thyroid peroxidase that traps oxidized iodide, preventing its use by thyroglobulin for thyroid hormone synthesis. Methimazole (0.4 mg/kg) inhibits the absorption of radiolabeled iodide by the thyroid gland in rats by 80.9%.3 It reduces the incidence of lymphocytic thyroiditis in the insulin-dependent type 1 diabetic BB/W rat. Methimazole has been used to induce hypothyroidism in mice. Formulations containing methimazole have been used in the treatment of hyperthyroidism.

Methimazole is a thyreostatic compound, and an antihormone, which is widely used in medicine for the treatment of hyperthyroidism.

Methimazole is a thioamide inhibitor of the enzyme thyroid peroxidase (TPO), with antithyroid activity. Upon administration, methimazole inhibits the metabolism of iodide and the iodination of tyrosine residues in the thyroid hormone precursor thyroglobulin by TPO; this prevents the synthesis of the thyroid hormones triiodothyronine (T3) and thyroxine (T4).

Methimazole is an antithyroid medication which is now considered the first line agent for medical therapy of hyperthyroidism and Graves disease. Methimazole has been linked to serum aminotransferase elevations during therapy as well as to a clinically apparent, idiosyncratic liver injury that is typically cholestatic and self-limited in course.
Methimazole, also known as tapazole or danantizol, belongs to the class of organic compounds known as imidazolethiones. These are aromatic compounds containing an imidazole ring which bears a thioketone group. Methimazole is a drug which is used for the treatment of hyperthyroidism, goiter, graves disease and psoriasis. Methimazole is soluble (in water) and a very weakly acidic compound (based on its pKa). Methimazole has been detected in multiple biofluids, such as urine and blood. Methimazole can be converted into methimazole S-oxide., 60-56-0.
