Fractions containing the desired product were combined and dried via centrifugal evaporation to give (R/5)-N- ((lH-benzo[d]imidazol-2-yl)methyl)-2-hydroxy-2-(4-(5-(3-phenyl-4- (trifluoromethyl)isoxazol-5-yl)-l,2,4-oxadiazol-3-yl)phenyl)acetamide (12.8 mg, 0.023 mmol, 49.3 % yield): LCMS = 561.1 [M+H]+; ‘H NMR (400 MHz, methanol- cU) 5 ppm 8.19 (2 H, d, J=8.58 Hz), 7.51-7.79 (10 H, m), 7.34 (2 H, dd, J=6.05, 3.19 Hz), 5.27 (1 H, s), 4.76-4.94 (2 H, m); HPLC peak RT = 2.6 min (Method E).

The residue was dissolved in EtOAc and washed with aq. K2CO3 solution followed by brine. The organic layer was then dried over Na2SO4, filtered, and concentrated in vacuo. The residue was purified by silica gel column chromatography eluting with 1-5 methanol/DCM v/v.

This intermediate (175 mg, 0.5 mmol) is dissolved in anhydrous THF (10 mL) and PPh3 (330 mg, 0.5 mmol) and 4-pentynol (46 muL, 0.5 mmol) are added. Then, after cooling to 0 C., DIAD (100 muL, 0.5 mmol) is slowly added, and after 15 min at 0 C., the mixture is heated under microwave irradiation at 110 C. for 1 h. Following flash chromatography purification (5:1 EtOAc-petr. et.), compound 10 is obtained in 64% yield. 1H NMR (CDCl3, 200 MHz) delta 7.95-7.90 (m, 1H), 7.71-7.67 (m, 1H), 7.34-7.26 (m, 2H), 5.79 (d, 2H), 2.30-2.20 (m, 2H), 2.03 (s, 1H), 1.95-1.92 (m, 2H), 1.80 (t, 2H), 1.71 (s, 9H), 1.47 (s, 9H) ppm.
