There was a good linear relationship in the exptl. concentrations of ethanol, acetone, isopropanol, acetonitrile, dichloromethane, Me tert-Bu ether, toluene (r≳0.9990). The average recoveries were ranged from 89.7% to 104.6%. The limit of detection were 1.40, 0.39, 1.60, 1.10, 0.74, 0.04 and 0.16 mg·L-1, resp. The method was sensitive, accurate and reproducible. It could be used for the detection of residual solvents in esomeprazole sodium.

A reversed-phase high-performance liquid chromatog. (RP-HPLC) method was established for the determination of related substances of esomeprazole sodium. The determination was performed on a Venusil XBP C18 (L) column (250 mm × 4.6 mm, 5 μm), with a mobile phase consisting of a mixture of acetonitrile and phosphate buffer (pH 7.6) by gradient elution. The column temperature was set at 30°C with a flow rate of 0.8 mL·min-1, and the detective wavelength was 302 nm. The related substances were completely separated from the main constituent. The calibration curves for 5-methoxy-2-mercaptobenzimidazole, omeprazole sulfone N-oxide, omeprazole sulfone, esomeprazole sodium and omeprazole sulfide revealed good linearities over the ranges of 3.99-29.94 μg·mL-1 (r=0.9998), 4.06-30.45 μg·mL-1 (r=0.9999), 2.46-18.42 μg·mL-1 (r=0.9998), 3.96-29.67 μg·mL-1 (r=0.9994), and 4.08-31.32 μg·mL-1 (r=0.9998), resp. The recoveries of the above compounds were 99.5% with RSD of 0.6%, 99.1% with RSD of 0.3%, 99.0% with RSD of 0.6%, 99.2% with RSD of 0.9%, and 99.0% with RSD of 0.7%, resp. The method could be used for testing the product quality of esomeprazole sodium.

NMR was employed to characterize the hydrogen-deuterium exchange for the proton pump inhibitors pantoprazole sodium, esomeprazole sodium, rabeprazole sodium, lansoprazole, and ilaprazole in methanol-d4. The results showed that the 1H and 13C signals of 8-CH2 (labeled as H8a, H8b, and C8) next to the sulfinyl (S=O) group of the proton pump inhibitors decreased significantly. Electrospray ionization mass spectrometry showed the presence of deuterated products. Basing on the NMR data, H8a and H8b of proton pump inhibitors were more active for the hydrogen-deuterium exchange. The most significant parameter governing the exchange was the salt-formation of the benzimidazole group. Therefore, the influence of salt-formation on hydrogen-deuterium exchange was characterized for ilaprazole; replacement of NH by N-Na on the benzimidazole group accelerated the exchange process. Tandem mass spectrometry showed that CH=S-OH was formed by tautomerism involving sulfinyl and CH2 groups in methanol-d4.

The compound was prepared by dissolving 0.185 g of 1.36 mmol Zn chloride in a 25 mL of H2O.
